Very short. Like 15 min. I forgot how frequent stops, but I think it's going on for 3 or 4.
I was traveling on bicycle and took a trolley from Old town San Diego to Santa Fe Station which be near the leading Amtrack Station, it only took similar to 10 min. This isn't exactly Downtown, but pretty close.
15-30 minutes, depending on traffic.
And I forget the number of stops. There's at smallest one at Mission Valley that I can remember. It's been awhile. :)

We only just did that trolley route on July 4th and it took 10 to 15 minutes from the Depot.
If you're coming from the Gaslamp, Old Town would be the 7th stop. Coming from Santa Fe Depot, Old Town is the 4th stop.
